DESCRIPTION:Professor Cullen Buie\n\nAbstract: \n \nIn this talk we will di
scuss work in our laboratory to use electric fields coupled with fluid flo
w in microfluidic devices to enable high throughput delivery of nucleic ac
ids to cells. Conventional electroporation approaches for bacterial gene d
elivery have distinct advantages\, but they are typically limited to relat
ively small sample volumes\, reducing their utility for applications requi
ring high throughput such as the generation of mutant libraries. Here\, we
present a scalable\, large-scale bacterial gene delivery approach enabled
by a disposable\, user-friendly microfluidic electroporation device requi
ring minimal fabrication. We demonstrate that the proposed device can outp
erform conventional cuvettes in a range of situations\, including across E
scherichia coli strains with a range of electroporation efficiencies\, and
we use its large-volume bacterial electroporation capability to generate
a library of transposon mutants in the anaerobic gut commensal Bifidobacte
rium longum. Results of this work hold exciting promise for accelerating g
enetic engineering of bacteria with wide ranging applications in industry
and healthcare. Further\, we will present recent efforts by a company spun
out of the Buie Laboratory\, Kytopen\, which is leveraging the electropor
ation work to enable scalable non-viral transfection of mammalian cells. A
pplications of this work include engineered cell therapies such as CAR-T\,
which are currently plagued by high costs and manufacturing issues. The n
on-viral transfection approach developed by Kytopen has the potential to s
implify and accelerate the development of life saving cellular therapies.
